I still have my original first edition hardcover of MARVEL MASTERWORKS: DOCTOR STRANGE Volume One from 1992 (which I have no intention of selling), but I'm considering getting the softcover version with the upgraded art reproduction to have as a reading copy, even though I dislike paperbacks in general.

How many printings have there been, and if more than one, does the paper quality differ between printings?

I don't know about Dr Strange. But I had the softcover of MMW Avengers 1, had an accident with it (put it on top of my laptop, fell asleep, woke up a couple of hours later and the thing was darn hot, ruined the cover) so bought it again this year. It's a second printing it seems (2012), it looks pretty much the same but the paper is a little less glossy and shiny than the paper they used in the first MMW softcovers. But not as bad as the paper they used in Thor 2 or FF 6 last year. But it's ok for me, besides of that the color and reproduction line is perfect.
