Legion Lost - Yes.  As long as you've read enough Legion to be familiar with the characters involved you'll be fine.  The book picks up mid-story, but the beginnings of that story happened off panel between the last old DCU issues and this one, so you're not missing out on anything by not having read the pre-Flashpoint Levitz/Johns Legion books - other than why Gates and Yera are on the team. 

Legion of Super-Heroes - Yes and No. 

Yes, in that there's no plots or sub-plots being carried over from the pre-relaunch series.  Virtually everything that Levtiz had going (LSV, Blue Entiity, return of Starman, Green Lantern Corps, Earth Man, Legion Academy) were all ended in August.  The only sub-plots that I can think of that are still hanging out there is Mysa/Mordru, the return of Karate Kid, and Brainy's obsession with cockroaches.  But all those were very minor bits in the pre-relaunch run and didn't figure at all in the new #1. 

But no, in the fact that it picks up right where the old series left off.  I think you'll get a LOT more out of the book if you've read the previous run.  But with a few minutes of research online you'll be able to figure out the basics of what happened before and be able to jump in with this book and not feel like you're in the middle of an existing story arc. This is probably the best jumping on point since Johns' Action run IMO.
