Well, in the early Weisinger columns (circa 1958), there were definitely plants - a couple were even addressed from his children, if I'm not mistaken, so they weren't disguised very well!

However, I do think that as time went on and he built up an inventory of letters, the more likely scenario to me would be that he would put letters aside that he thought would make good story material, and then publish them later to coincide with that story's release. There certainly were many instances where exchanges in his LC led to a tale being written (Luthor's origin being one of them!).

One other funny aside about this - strange but true - I remember another message board where someone offered up an example of what they thought was a Weisinger "plant", and said how could anyone believe these letters were real, when lo and behold, one of the regular members of that community stepped forward and said, "wait a minute, that was me - I wrote that letter!"
